There are two main reasons to get video to play in Quicktime.
1) To make your own DVDs with iDVD.
2) To convert videos to a format that plays on iPods (.m4v). 
I do a lot of 1, and with my best friend giving me an iPod as a wedding present I've started to do some of 2.  Previously I would load the video in VLC and have it tell me what codecs were being used.  I'd then try and find a version of those codecs for Quicktime.  It was ususally hit or miss.
There is a better way.  You just install an open-source program called Perian that installs Quicktime support for lots of common video formats.  It works perfectly!
